Abstract

Mammalian AlkB homologues (ABH) contain a 2-oxoglutarate and iron dependent dioxygenase domain, placing these proteins in the 2OG/Fe2+ dependent dioxygenase superfamily. The mammalian ABH2 and ABH3 proteins are oxidative demethylases which catalyze the removal of alkylation damage from dsDNA, ssDNA and RNA with different efficiencies. Six other mammalian homologues have been identified with possible roles in DNA, RNA, and protein demethylation. Here, a study was initiated to characterize the in vivo function of the murine homologue mAbh7. With this aim in mind a conditional mAbh7 knockout mouse was created. Additionally, the mRNA expression profiles of wild-type mAbh7 in organs and embryos were elucidated. The similarity between the human homologue hABH7 and the mouse homologue mAbh7 provides a rational to suggest a similar function for these two proteins. With these homologies in mind, an attempt was made to verify hABH7 protein interaction partners identified using a yeast 2-hybrid screen.
